| 目的:建立党参中总氨基酸含量的测定方法。方法:收集12批不同产地的党参。以L-精氨酸为对照品,采用茚三酮显色体系,建立用紫外可见分光光度法(UV-VIS)测定党参中总氨基酸含量方法。结果:确定茚三酮比色法的最佳反应条件为:缓冲液pH7.2,用量0.5mL,茚三酮显色剂用量0.8mL,沸水浴加热15min;L-精氨酸在7.3~16.9μg/mL范围内与吸光度线性关系良好,回归方程为A=77.155c-0.293(R2=0.9995,n=6),平均加样回收率为99.4%(RSD=2.0%,n=6),12批党参中总氨基酸含量为27.0~54.2mg/g。结论:本研究建立的方法操作简单、专属性强、重复性良好,可用于党参药材中总氨基酸的含量测定。 |

| Objective Establish a method for determining total amino acids in Codonopsis pilosula. Methods Collect 12 batches of Codonopsis pilosula from different origins. The UV - VIS method for the determination of total amino acids content was establishedby the ninhydrin colorimetry with arginine as the reference. Results The optimal reaction conditions for the ninhydrin colorimetry colorimetric method are a buffer solution with an optimal pH value of 7 . 2 and a dosage of 0. 5 mL; the dosage of ninhydrin colorimetry chromogenic agent is 0. 8 mL , the heating in a boiling water bath for 15 minutes. The linear range of arginine was 7. 3 ~ 16. 9 μg/mL , the linear re- gression equation was A = 77 . 155c - 0. 293 (R2 = 0. 9995 , n = 6) and the average recovery was 99. 4% ( RSD = 2. 0% , n = 6) . The contents of total amino acids in 12 bantches of Codonopsis pilosula were 27. 0 - 54. 2 mg/g. Conclusion This method is simple , specifically , reproducible. Itcan be used for the determination of total amino acid content in Codonopsis pilosula. |
