| 目的:建立马棘的质量标准,并初步探究马棘的抑菌作用。方法:对马棘进行性状、显微和薄层鉴别,采用UPLC法同时测定单咖啡酰酒石酸、龙胆酸和菊苣酸含量,参照2020年版《中国药典》四部通则方法测定水分、总灰分、浸出物含量;分别采用牛津杯法和二倍稀释法测定马棘茎、叶水提液对金黄色葡萄球菌、大肠杆菌、枯草芽孢杆菌、沙门杆菌、铜绿假单胞菌的抑菌圈直径和最小抑菌浓度。结果:药材粉末呈棕黄色至灰褐色,显微鉴别特征明显,薄层鉴别分离度好,斑点清晰。暂定水分、总灰分分别不得超过13%、9%,浸出物含量不得低于16%。单咖啡酰酒石酸、龙胆酸、菊苣酸在20~500mg/g范围内线性关系良好(r2≥0.9998)。马棘叶部位的抑菌圈直径在13~19mm之间,其对金黄色葡萄球菌为高敏,对铜绿假单胞菌、大肠杆菌为中敏;马棘茎部位的抑菌圈在9~13mm之间,其对铜绿假单胞菌、大肠杆菌为低敏至中敏;叶、茎水提物对沙门杆菌均为中敏、对枯草芽孢杆菌均无抑菌活性。表明叶提取物对4种菌株的抑菌效果强于茎提取物的抑菌效果。结论:该方法简便、准确、重复性好,可为马棘质量控制提供依据;马棘的叶、茎水提物对除了枯草芽孢杆菌以外的4种菌种有较好的抑菌活性。 |

| Objective To establish quality standards for Indigofera pseudotinctoria Matsum. (I. pseudotinctoria Matsum. ) and pre- liminary investigation of the bacteriostatic effect of Ip Matsum. Methods Ip Matsum. were subjected to morphological identification , microscopic identification , and thin - layer identification. The content of caftaric acid , 2 , 5 - dihydroxybenzoic acid and cichoric acid was simultaneously determined by UPLC , and the water content , total ash content , and extract content were detected according to the method stated in 2020 edition of Chinese Pharmacopoeia. The zone of inhibition and minimal inhibit concentration of the aqueous ex- tract from the stems and leaves of Ip Matsum. against Staphylococcus aureus , Escherichia coli , Bacillus subtilis , Salmonella , and Pseudomonas aeruginosa were determined by Oxford cup method and double dilution method respectively. Results The medicinal pow- der was brownish yellow to grayish brown in color , with obvious microscopic identification characteristics and showed good resolution and clear characteristic spots by TLC . It is tentatively stipulated that the water content and the total ash content should not exceed 13% and 9% respectively , and the extract content should not be less than 16% . The linear relationship between caftaric acid , 2 , 5 - di- hydroxybenzoic acid and cichoric acid is good in the range of 20 - 500 mg/g ( r2 ≥0. 999 8) . The diameter of the inhibitory zone in the Ip Matsum. leaves is between 13 - 19 mm , it is highly sensitive to Staphylococcus aureus and moderately sensitive to Pseudomonas aeruginosa and Escherichia coli; the diameter of the inhibitory zone in the Ip Matsum. stem is between 9 - 13 mm , it is low to moder- ately sensitive to Pseudomonas aeruginosa and Escherichia coli; the water extract of leaf and stem is moderately sensitive to Salmonella and has no antibacterial activity against Bacillus subtilis . The results showed that the antibacterial effect of leaves extract on the four strains was stronger than that of stem extract. Conclusion This method is simple , accurate , and has good repeatability , and can pro- vide a basis for quality control of Ip Matsum. The aqueous extracts from the leaves and stems of Ip Matsum. have good antibacterial ac- tivity against four species except Bacillus subtilis . |
