      目的:研究十一方药酒的抗炎作用及其谱效关系,并对十一方药酒安全性进行评价。方法:采用二甲苯致耳肿胀、角叉菜胶致足肿胀评价十一方药酒外用给药的抗炎作用。并以角叉菜胶致足肿胀组织中的前列腺素E2(PGE2)、组胺(HIS)、白介素-1β(IL-1β)含量,以及致炎足组织的病理变化探讨抗炎作用机制。HPLC法建立十一方药酒的指纹图谱,以二甲苯致耳肿胀度、角叉菜胶致足肿胀度为药效指标,采用灰色关联度法分析HPLC共有峰与药效指标间的谱效相关性;采用家兔皮肤刺激性试验及豚鼠皮肤过敏性试验对十一方药酒的安全性进行初步评价。结果:与模型组比较,十一方药酒高、中、低浓度组能明显抑制二甲苯造成的小鼠耳肿胀(P<0.01);十一方药酒高、中浓度组能明显抑制角叉菜胶造成的足肿胀(P<0.05或P<0.01),十一方药酒高浓度组能降低肿胀足组织炎症因子PGE2、HIS、IL-1β含量(P<0.05),HE染色显示十一方药酒各组能减少致炎部位的炎性细胞浸润;谱效相关性分析显示,十一方药酒指纹图谱中峰3(羟基红花黄色素A)、峰2、峰13等14个色谱峰为药效相关成分;该药酒对新西兰兔完整皮肤和破损皮肤无刺激性,对豚鼠无过敏性。结论:十一方药酒药效稳定且安全有效,可通过降低炎症因子含量、减少致炎部位的炎性细胞浸润发挥抗炎消肿作用。十一方药酒指纹图谱中有14个共有峰与其抗炎作用的关系密切,这些色谱峰所代表的成分可能是十一方药酒抗炎作用的有效成分群。

      Objective To study the anti - inflammatory effect and spectrum effect relationship of Shiyifang vinum , and evaluate the safety of Shiyifang vinum. Methods Xylene - induced ear swelling and carrageene - induced foot swelling were used to evaluate the anti - inflammatory effects of Shiyifang vinum. The anti - inflammation mechanism was investigated by the contents of prostaglandin E2 (PGE2) , histamine (HIS) and interleukin - 1β (IL - 1β) in swollen foot tissues induced by carrageenan and the pathological changes of foot tissues induced by inflammation. HPLC method was used to establish the fingerprint of Shiyifang vinum , taking the degree of ear swelling induced by xylene and the degree of foot swelling induced by carrageenan as the pharmacodynamic indexes . Grey corre- lation method was used to analyze the spectral correlation between HPLC common peak and pharmacodynamic indexes . The safety of Shiyifang vinum was evaluated by rabbit skin irritation test and guinea pig skin allergy test. Results Compared with model group , high , medium and low concentration groups of Shiyifang vinum could significantly inhibit the ear swelling caused by xylene ( P < 0. 01) . High and medium concentration groups of Shiyifang vinum could significantly inhibit the foot swelling caused by carrageenan (P < 0. 05 or P < 0. 01) , and high concentration group of Shiyifang vinum could reduce the contents of inflammatory factors PGE2 ,HIS and IL - 1βin swollen foot tissue (P < 0. 05) . HE staining showed that Shiyifang vinum could reduce the inflammatory cell infiltration in the inflammatory site in each group . The spectral correlation analysis showed that 14 chromatographic peaks , including peak 3 (hydroxyl safflower yellow A) , peak2 , peak13 , were pharmacodynamic related components in the fingerprint of Shiyifang vinum.The liquor was not irritating to intact and damaged skin of New Zealand rabbits , and was not allergic to guinea pigs . Conclusion The effect of Shiyifang vinum is stable , safe and effective , and it can play an anti - inflammatory and detumescence role by reducing the content of inflammatory factors and reducing the inflammatory cell infiltration in the inflammatory site . There are 14 common peaks in the fingerprint of Shiyifang vinum , which are closely related to its anti - inflammatory effect. The components represented by these chromatographic peaks may be the effective component group of Shiyifang vinum.
