目的:优选消渴健脾方中有效成分丹酚酸B的提取工艺。方法:以甲醇-5%乙酸(35∶65),柱温23℃,流速为1.0mL/min,检测波长为281nm,建立丹酚酸B的HPLC色谱条件;采用正交试验设计对加水量、煎煮时间、煎煮次数3个因素进行考察,优选消渴健脾方中有效成分丹酚酸B的提取工艺。结果:优选的最佳提取条件为加水量10倍、煎煮时间45min、煎煮次数2次。验证试验结果表明,在此方法下消渴健脾方中有效成分丹酚酸B的含量平均值为0.882mg/g。结论:本研究优选的提取工艺稳定、可靠,可用于消渴健脾方有效成分丹酚酸B的提取。 |

Objective To optimize the extraction process of salvianolic acid B,an active ingredient in Xiaoke Jianpi Formula.Methods The HPLC chromatographic conditions of salvianolic acid B were established using methanol - 5% acetic acid (35 ∶65),column temperature of 23 ℃ , flow rate of 1.0 mL/min,and detection wavelength of 281 nm. Three factors,including the amount of added water,the decoction time,and the number of times of decoction were investigated using orthogonal experimental design , to optimize the extraction process of salvianolic acid B . Results The optimal extraction process was 10 times of water addition ,45 min of decoction time and 2 times of decoction. The results of the validation test showed that the average content of salvianolic acid B in the formula was 0.882 mg/g. Conclusion The preferred extraction process is stable and feasible,and can be used for the extraction of salvianolic acid B . |
