目的:探究由农药高效氯氰菊酯菊(β-Cypermethrin,β-CYP)诱导建立的小鼠肝脏损伤模型中,菊苣乙醇提取物(CG-Ι)对农药所致肝脏损伤的保护作用。方法:80%乙醇对菊苣粉进行回流提取浸膏以备用;取60只清洁级小鼠随机分为正常对照组、CG-Ι低剂量组、CG-Ι中剂量组、CG-Ι高剂量组、农药组、中药组;正常对照组灌胃生理盐水、CG-Ι低剂量组灌胃CG-Ι(50mg/kg)+β-CYP(40mg/kg)、CG-Ι中剂量组灌胃CG-Ι(100mg/kg)+β-CYP(40mg/kg)、CG-Ι高剂量组灌胃CG-Ι(200mg/kg)+β-CYP(40mg/kg)、农药组灌胃β-CYP(40mg/kg)、中药组灌胃CG-Ι(100mg/kg);实验期间观察小鼠毛发、精神状态等基本情况并记录体重;持续4周后,收集小鼠肝脏并称重,对肝组织进行苏木精-伊红(hematoxylin-eosin,HE)染色观察小鼠肝脏组织结构;同时检测小鼠肝组织匀浆中氧化损伤指标谷丙转氨酶(ALT)、天冬氨酸氨基转移酶(AST)、超氧化物歧化酶(SOD)、丙二醇(MDA)水平。结果:随着实验的进行农药组与低剂量组小鼠毛发变得混乱,精神状态低迷;除低剂量组和农药组小鼠体重增长缓慢外,其余各组小鼠体重增长正常;低剂量组和农药组小鼠肝脏器系数相较于其余各组出现异常;低剂量组和农药组小鼠ALT、AST表达水平较高,而中、高剂量组和中药组小鼠表达水平较低与正常对照组相近;低剂量组和农药组小鼠SOD活力较低,而中、高剂量组和中药组小鼠SOD活力较高且与正常对照组相近;低剂量组和农药组小鼠MDA表达水平较高,而中、高剂量组和中药组小鼠MDA表达水平较低与正常对照组相差较小。肝脏病理切片结果显示中、高剂量CG-Ι对β-CYP所诱导的小鼠肝脏组织病理学变化有良好的治疗效果。结论:与正常对照组相比较,低剂量CG-Ι对小鼠ALT、AST、SOD、MDA水平的调控无显著作用且体重增长缓慢,脏器系数异常,因而对肝脏的保护作用不明显;而中、高剂量CG-Ι对小鼠ALT、AST、SOD、MDA水平的调控有显著影响且组体重增长、脏器系数和肝组织结构均正常,其中高剂量组最接近正常对照组,因此中、高剂量CG-Ι对β-CYP所致小鼠肝脏损伤具有较好的保护作用,机制可能是CG-Ι能增强肝细胞的抗氧化损伤能力。 |

Objective To investigate the protective effect of chicory ethanol extract (CG-Ι) on pesticide - induced liver injury in a mouse liver injury model induced by the pesticide cypermethrin ( β- Cypermethrin , β- CYP) . Methods Chicory powder was re- fluxed with 80% ethanol to extract for later use; Sixty clean - grade mice were randomly divided into normal control group , CG - Ιlow - dose group , CG - Ιmedium - dose group , CG - Ιhigh - dose group , pesticide group and traditional Chinese medicine group; The normal control group was given normal saline by gavage; CG - Ιlow - dose group was given CG - Ι( 50 mg/kg) + β- CYP (40 mg/ kg) ; CG - Ιmedium - dose group were given CG - Ι ( 100 mg/kg) + β- CYP (40 mg/kg) ; The high - dose CG - Ιgroup was given CG - Ι(200 mg/kg) + β- CYP (40 mg/kg) ; The pesticide group was given β- CYP(40 mg/kg) , and the traditional Chi- nese medicine group was given CG - Ι( 100 mg/kg) ; During the experiment , the basic conditions such as the hair and mental state of the mice were observed , and the body weight was recorded ; After 4 weeks of experimentation , the livers of mice were weighed , and the liver tissues were stained with hematoxylin - eosin (HE) to observe the structure of mouse liver tissues , and the levels of alanineaminotransferase (ALT) , aspartate aminotransferase (AST) , superoxide dismutase (SOD) and propylene glycol (MDA) , which are indicators of oxidative damage in the homogenates of mouse liver tissues , were detected. Results Mice in the pesticide group and the low - dose group had chaotic hair and low mental state in the later stage; Except for the low - dose group and the pesticide group , the weight gain of the mice in the other groups was normal; Compared with the other groups , the liver organ coefficient of mice in the low - dose group and the pesticide group was abnormal; The expression levels of ALT and AST in the low - dose group and pesticide group were higher , and the levels of ALT and AST in the medium - dose and high - dose groups and the traditional Chinese medicine group were lower than those in the normal control group; The SOD activity of mice in the low - dose group and the pesticide group was lower , and the SOD activity in the medium - dose and high - dose groups and the traditional Chinese medicine group was higher and similar to that of the normal control group; The expression level of MDA in mice in the low - dose group and pesticide group was higher , and the expression level of MDA in mice in the medium - dose and high - dose groups and traditional Chinese medicine group was lower , which was slightly different from that in the normal control group . The results of liver pathological sections showed that medium and high doses
of CG - Ιhad a good therapeutic effect on the histopathological changes in the liver induced by β- CYP in mice . Conclusion Com- pared with the normal control group , low - dose CG - Ιhad no significant effect on the regulation of ALT , AST , SOD , MDA levels in mice , and the weight gain was slow , and the organ coefficient was higher , so the protective effect on the liver was not obvious , while the medium and high doses of CG -Ιhad a significant effect on the regulation of ALT , AST , SOD , and MDA levels in mice , and the weight gain , organ coefficient and liver tissue structure of the group were normal.,and the high - dose group was closest to the normal control group . High - dose CG - Ιhas a good protective effect on liver injury induced by β- CYP in mice , and the mechanism may be that CG - Ιcan enhance the antioxidant damage ability of hepatocytes . |
