目的:探究离蕊金花茶对急性肝损伤小鼠的保护作用及体外抗氧化活性。方法:采用 DPPH 自由基清除实验探讨离蕊金花茶的体外抗氧化活性;通过四氯化碳(CCl4 )构建小鼠急性肝损伤模型,评价离蕊金花茶对急性肝损伤的影响。将昆明种雄性小鼠60只随机分为6个组:空白组、模型组、护肝片组(0.55 g/kg)、离蕊金花茶低 (5 .2 g/kg) 、中(10.4 g/kg)、高(20.8g/kg)剂量组 ,每天给药一次 ,连续 14 d,最后一次给药1h后,以 0. 1% CCl4橄榄油溶液建 立急性肝损伤模型。16 h 后取血、取肝脏低温保存备用。检测血清中γ- 谷氨酰转肽酶 (γ- GT)、天冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)、丙氨酸氨基转移酶 (ALT) 和乳酸脱氢酶 ( LDH) 水平及肝脏组织中丙二醛 ( MDA) 和谷 胱甘肽(GSH)含量;制作肝脏病理切片,观察肝脏组织的病理变化。结果:离蕊金花茶具有较好的体外抗氧化活性;能降低小鼠血清中 ALT、AST 、γ- GT、ALP 和 LDH 水平;能使小鼠肝脏组织中 MDA 含量降低以及升高 GSH 含量。结论:离蕊金花茶具有较好的抗氧化活性,对急性肝损伤具有一定的保护作用。 |

Objective To investigate the protective effect and antioxidant activity of Camellia liberofilamenta on mice with acute liver injury.Methods DPPH free radical scavenging experiment was used to investigate the antioxidant activity of Camellia liberofila- menta in vitro.The mouse model of acute liver injury was established by carbon tetrachloride (CCl4 ) to evaluate the effect of Camellia liberofilamenta on acute liver injury.Sixty male mice of Kunming species were randomly divided into 6 groups :Blank group , model group ,Hugan tablet group (0. 55 g/kg),low (5.2 g/kg) ,medium(10. 4 g/kg) and high(20. 8 g/kg) dose groups were adminis- tered once a day for 14 consecutive days ,and 0. 1% CCl4 olive oil solution was used to establish acute liver injury model after the lastadministration for 1 hour.16hlater ,blood and liver were taken and stored at low temperature for later use .The levels of γ- glutamyl-transpeptidase (γ- GT) ,aspartate aminotransferase( AST) ,alkaline phosphatase( ALP) ,alanine aminotransferase ( ALT) and lactate dehydrogenase (LDH) in serum and the contents of malondialdehyde (MDA)and glutathione (GSH)in liver were detected. The pathological sections of liver were prepared to observe the pathological changes of liver tissue.Results Camellia liberofilamenta had good antioxidant activity in vitro.It can reduce the levels of ALT ,AST ,γ- GT ,ALP and LDH in serum of mice.It can decrease MDA content and increase GSH content in liver tissue of mice .Conclusion Camellia liberofilamenta has good antioxidant activity and protective effect on acute liver injury. |
