      目的:对蒙自藜芦总生物碱提取工艺进行优化。方法:以蒙自藜芦总生物碱含量及3个成分藜芦碱、藜芦胺、介芬胺为指标,通过静态吸附和动态吸附实验考查上样体积、上样液质量浓度、径高比、上样液PH值、水洗用量、洗脱剂及其用量,优化出最佳纯化工艺条件。结果:选用D101型大孔树脂纯化蒙自藜芦总生物碱提取物,上样体积50mL,径高比1:5,上样液总生物碱质量浓度为0.2mg/mL,上样液pH值为9,水洗用量为4BV,洗脱剂为80%乙醇,洗脱剂用量为8BV,纯化后的蒙自藜芦总生物碱转移率大于60%,含量为62.88%,比纯化前提高5.74倍。结论:D101型大孔树脂纯化蒙自藜芦总生物碱工艺稳定可行,可为后续制剂开发提供参考依据。

      Objective In Veratrum mengtzeanum total alkaloid extraction technology was optimized. Methods Taking the total al-kaloid content and three components veratrine, veratrine, and mesofenamine as indicators, the sample volume, mass concentration ofsample solution, diameter/height ratio, pH value of sample solution, amount of water washing, eluent and its amount of sample solu-tion were examined by static adsorption and dynamic adsorption experiments, and the best purification process conditions were opti-mized. Results D101 macroporous resin was used to purify the total alkaloid extract of Veratrum mengtzeanum. The sample volume was50mL and the diameter/height ratio was 1:5, on the quality of samples, total alkaloids concentration is 0.2 mg/mL, pH value of thesample fluid 9, water dosage of 4 bv, eluent is 80% ethanol, dosage of eluent is 8 bv, purified of Veratrum mengtzeanum transfer ratemore than 60% of total alkaloid content is 62. 88% , and 5. 74 times higher than that of before purification. Conclusion D101 macro-porous resin purification process is stable and feasible, which can provide a reference for the subsequent preparation development.
