      目的:研究并建立半夏药材HPLC特征图谱,提高其质量控制水平。方法:采用SVEAC1sOpal色谱柱(4.6 mmx250 mm,5μm),以甲醇一水作为流动相进行梯度洗脱,流速为0.9mL/min,柱温为30℃,检测波长为270nm,建立了半夏药材的指纹图谱,运用国家药典委员会《中药色谱指纹图谱相似度评价系统(2012版)》对测定的17批半夏药材数据进行处理分析。结果:建立了以9个特征峰为指标成分的半夏药材HPLC特征图谱,以鸟苷色谱峰为参照,其他8个特征峰的相对保留时间分别为(0.503±0.050)min、(0.563±0.056)min、(0.643±0.064)min、(0.928±0.093)min、(1.490±0.149)min、(1.536±0.154)min、(2.171±0.217)min、(2.210±0.221)min。结论:建立了同时测定不同来源半夏药材化学成分的HPLC特征图谱方法,该方法经过方法学验证,结果可靠,可用于半夏药材的质量控制。

      Objective To study and establish the specific chromatogram of Pinelliae Rhizoma by HPLC, so as to improve its quali-ty control. Methods SVEA C8 Opal chromatographic column (250 mm ×4. 6 mm, 5 um) was used for gradient elution with methanol-water as the mobile phase at the flow rate of 0.9 mL/min. The column temperature was 30 T and the detection wavelength was270 nm. The fingerprint of Pinelliae Rhizoma was established. The data of 17 batches of pinellia pinellia were processed and analyzedby the Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System (2012 Edition) of the Chinese Pharmacopoeia Com-mission. Results Nine common peaks were found through the similarity evaluation system. With phenylalanine as the reference, therelative retention time of the other eight peaks were (0. 503 ±0. 050) min, (0. 563 ±0. 056) min, (0. 643 ±0. 064) min, (0. 928±0. 093) min, (1. 490 ±0. 149) min, (1. 536 ±0. 154) min, (2. 171 ±0. 217) min, (2. 210 ±0. 221) min, respectively.Conclusion HPLC specific chromatogram method for simultaneous determination of chemical components of Pinelliae Rhizoma from dif-ferent sources is established. The method is proved to be reliable and can be used for quality control of Pinelliae Rhizoma.
