      目的:建立HPLC法同时测定美洲大蠊标准汤剂中尿嘧啶、次黄嘌呤、尿苷、肌苷、鸟苷、原儿茶酸的含有量。方法:采用WATERS Symmetry Shield C1g色谱柱(250mm×4.6mm,5um);甲醇-0.1%磷酸水作为流动相,梯度洗脱;体积流量0.6mL/min;检测波长254nm;柱温20℃。结果:测定显示尿嘧啶、次黄嘌呤、尿苷、肌苷、鸟苷、原儿茶酸分别在0.041~0.341 μg(r=0.9994)、0.097~0.810μg(r=0.9994)、0.020~0.168 μg(r=0.9999)、0.277~2.308 μg(r=0.9999)、0.016~0.135μg(r=0.9997)、0.131~1.096μg(r=0.9998)范围内线性关系良好,平均加样回收率分别为99.93% (RSD =1.18%)、 98.03% (RSD =1.05%)、 97.17% (RSD =1.29%)、 100.19% (RSD =1.91%)、98.94% (RSD=1.11%)、100.70%(RSD=1.05%)。十批美洲大蠊标准汤剂各成分含量范围分别为:尿嘧啶0.14~0.21mg/g、次黄嘌呤0.22~0.36mg/g、尿苷0.06~0.08mg/g、肌苷2.17~3.15mg/g、鸟苷0.03~0.07mg/g、原儿茶酸0.35~0.56mg/g,处于均值的±30%范围内。结论:美洲大蠊标准汤剂含量测定方法准确、可靠,可用于美洲大蠊配方颗粒的质量参考。

      Objective To establish the HPLC method for simultaneously determining the contents of uracil, hypoxanthine, uri-dine, inosine, guanosine, protocatechuic acid in standard decoction of Periplaneta americana. Methods The analysis was performedon WATERS Symmetry Shield Cg column (250 mm ×4.6 mm, 5 um) . The chromatograhic separation was gradient by methanal –0. 1% phosphoric acid; The flow rate was 0. 6mL/min. The column temperature was 20 T and the detection wavelength was set at254 nm. Results The results showed that Uracil, hypoxanthine, uridine, inosine, guanosine, protocatechuic acid showed good line-ar relationships within the ranges of 0.041 –0. 341 ug (r =0. 9994), 0.097 – 0. 810 ug (r =0. 9994), 0.020 – 0. 168 μg (r =0. 9999), 0. 277 –2.308 μg (r =0. 9999), 0.016 –0. 135 μg (r =0. 9997), 0. 131 - 1.096 μg (r =0. 9998) and their averagerecoveries were 99. 93% (RSD = 1. 18% )、 98. 03% (RSD = 1. 05%)、 97. 17% (RSD = 1. 29% )、 100. 19% (RSD =1. 91%)、 98. 94% (RSD =1. 11%)、 100. 70% (RSD =1. 05%), respectively. The content ranges of each component in ten bat-ches of Standard Decotion of Periplaneta americana were as follows: 0. 14 –0. 21 mg/g of uracil, 0. 22 –0.36 mg/g of hypoxanthine,0. 06 –0. 08 mg/g of uridine, 2. 17 –3. 15 mg/g of inosine, 0. 03 – 0. 07 mg/g of guanosine, 0. 35 – 0. 56 mg/g of protocatechuicacid. The content ranges were in the range of the mean ±30%. Conclusion The method is simple and accurate, which can be usedfor the quality control of Periplaneta americana dispensing granules.
