      【摘 要】目的:观察升降散对糖尿病大鼠双歧杆菌、大肠杆菌及IL-6的影响,探讨其作用机制。方法:取70只SD雄性大鼠,随机取10只作为正常对照组,余60只用高脂饲料喂养配合小剂量链脲佐菌素诱导建立糖尿病模型,取成模大鼠50只分为模型对照组、拜糖平组、升降散低剂量组、升降散中剂量组和升降散高剂量组,共分6组,每组10只,各组按相应药物剂量干预6周后,每组取8只大鼠样本采用Elisa法检测IL-6、INS,RealtimePCR技术进行细菌定量测定。结果:治疗前,模型对照组、拜糖平组、升降散低、中、高三个剂量组较正常对照组空腹血糖(FBG)、IL-6、INS及大肠杆菌数量均明显升高,但双歧杆菌数量明显减少;治疗前后,正常对照组及模型对照组组内各指标无明显变化。治疗后,拜糖平组及升降散低、中、高剂量FBG在2周、4周及6周均逐渐降低,且高于同时间正常对照组,低于同时间模型对照组(P<0.05);IL-6、INS、大肠杆菌数量较模型对照组明显减少,但同时高于正常对照组(P<0.05);双歧杆菌数量较模型对照组、正常对照组明显增多(P<0.01)。尤以升降散中剂量组增加双歧杆菌数量、降低大肠杆菌数量,抑制炎症因子、降低胰岛素含量效果明显。结论:升降散能通过增加益生菌数量,降低致病菌数量达到调整肠道菌群结构的目的,从而抑制炎症状态,降低血糖。其部分作用机制可能是通过调整肠道菌群、抑制炎症因子而减轻胰岛素抵抗(IR),增加胰岛素利用率实现的。

      Abstract:ObjectiveToobservetheeffectandthemechanismofChineseherbalmedicineonBifidobacterium,EscherichiacoliandIL-6indiabeticrats.Methods70SDmalerats,10ratsasnormalcontrolgroup,onlymorethan60highfatdietcombinedwithlowdoseofstreptozotocininduceddiabeticmodelwasestablished,themodel50ratsweredividedintomodelgroup,BaiTangpinggroup,Shengjiangsanlowdosegroup,middledosegroupShengjiangsanShengjiangsanandhighdosegroupsweredividedinto6groups,10ratsineachgroup,eachgroupaccordingtothecorrespondingdoseofdrugfor6weeks,8ratsfromeachgroupofsampleswasdetectedbyElisaIL-6,INSRealtime,PCRtechnologyforquantitativedeterminationofbacteria.ResultsBeforetreatment,modelgroupandacar bosegroup,Shengjiangsanlow,mediumandhighdosegroupscomparedwithnormalcontrolgroup,fastingbloodglucose(FBG),IL-6,INSandthenumberofEscherichiacoliweresignificantlyincreased,butthenumberofbifidobacteriawassignificantlyreduced;beforeandafterthetreatment,thenormalcontrolgroupandmodelcontrolgroupeachindexinthegrouphadnoobviouschange.Aftertreatment,acarboseShengjiangsangroupandlow,middleandhighdoseofFBGin2weeks,4weeksand6weeksweregraduallydecreased,andhigherthanthatofthenormalcontrolgroup,comparedwithmodelcontrolgroup(P<0.05);IL-6,INS,thenumberofEscherichiacolithanthemodelcontrolgroupwassignificantlyreduced,butatthesametimehigherthanthenormalcontrolgroup(P<0.05);thenumberofbifidobacteriacomparedwithmodelcontrolgroup,normalcontrolgroupincreasedsignificantly(P<0.01)Especiallyinthemiddledosegroup,thenumberofbifidobacteriaincreased,thenumberofEcolidecreased,theinflammatoryfactorsandinsulincontentweredecreasedsignificantly.ConclusionByincreasingthenumberofprobioticbacteria,reducingthenumberofpathogenstoadjustthestructureofintestinalflora,therebyinhibitingtheinflammatorystate,reducingbloodglucose.Partofitsmechanismmaybetoreduceinsulinresistance(IR)andincreaseinsulinutilizationbyregulatingintestinalfloraandinhibitinginflammatoryfactors.
